Alzheon was founded in 2013 by Martin Tolar, a physician-scientist, building on research originally developed at Harvard Medical School and Massachusetts General Hospital into a molecule intended to selectively stabilize toxic amyloid beta oligomers, one of the proteins widely implicated in the buildup of plaques associated with Alzheimer's disease. The company's lead candidate, an oral small-molecule drug marketed under the name ALZ-801, is designed as a prodrug of tramiprosate, an earlier compound studied in Alzheimer's research, reformulated to improve its pharmacological properties and specifically targeted at patients who carry certain genetic risk variants, such as two copies of the APOE4 gene, that put them at especially high risk of developing the disease. Alzheon has positioned ALZ-801 as a potential oral alternative to the antibody infusion therapies, such as those developed by Biogen and Eisai or Eli Lilly, that have recently reached the Alzheimer's drug market but require regular infusions and carry certain safety monitoring requirements related to brain swelling and bleeding. As an oral pill, Alzheon's candidate would in principle be easier for patients to take regularly and potentially more scalable for widespread use if its clinical trial results hold up. The company has run Phase 3 clinical trials of ALZ-801 in genetically defined patient populations and has published data suggesting a slowing of cognitive decline alongside a favorable safety profile relative to antibody therapies. Alzheon has raised funding from investors including Alzheon's own founders and outside biotech investors, and continues to advance its regulatory filings with the U.S. Food and Drug Administration as it seeks approval, competing in a rapidly evolving Alzheimer's treatment landscape that now includes several approved and late-stage therapies from much larger pharmaceutical companies.
